Actually, when you isolate a variety of life stages from a colony with a queen, you may sometimes grab secondary reproductives or individuals that can be transformed into new secondary reproductives. In most termite colonies aside from the main queen there's also a good amount of secondary reproductives, though in this case I think a worker or a younger individual that hadn't differentiated yet became a secondary reproductive.
